About this structure

The human heat shock-70 like protein-1 (HSPA1L; also known as Hsp70-hom or Hsp70t) is a cytosolic member of the Hsp70 family with high levels of expression in testis (1-3). By analogy with other Hsp70 proteins, HSPA1L stabilizes existing proteins against aggregation and mediates the folding of newly translated proteins. Polymorphisms in the gene have been linked to human disease, predominantly in the region coding for the C-terminal peptide binding domain.

We have previously determined the crystal structure of the ATPase domain of the HSPA6 protein (PDB: 3FE1). Here, the structure of the corresponding part of HSPA1L (residues M6 – K386) at 2.2 Å resolution is presented. Our structure shows the protein with one manganese ion and the products of ATP hydrolysis (one molecule of ADP and one phosphate group) bound in the cleft between the two major lobes of the ATPase domain. Coordinates and structure factors have been deposited in the Protein Data Bank under PDB accession code 3GDQ.
